Video 7.2. Contrast-enhanced μCT (sagittal view) of a mouse knee following non-invasive ACL injury as previously described (Christiansen et al., Osteoarthritis and Cartilage, 2012). Mice were sacrificed immediately after injury (or sham injury) and knees were removed and stained with phosphotungstic acid (PTA; 0.3% in 70% ethanol) for one week before being scanned with μCT (2 μm nominal voxel size). PTA binds to fibrin and collagen, proteins that are ubiquitous in connective tissue. Therefore, staining with PTA allows for μCT imaging of musculoskeletal soft tissues including tendon, ligament, articular cartilage, meniscus, muscle, and synovium.
